Recently, a phase II clinical trial of the MSL for multiple sclerosis was started in New Zealand. The trial was developed with the aim of utilizing the proven safety of MSLs and the benefits of monotherapy in the preclinical mouse model for multiple sclerosis. Preliminary data from the study indicates that as expected, the MSL is well tolerated at the dose levels used in the trial, and is able to reach the target CNS tissues within 2hrs of systemic administration, with an approximately 50% reduction in target T-cells in the brain. It is also promising that the active MSL immunotoxin is able to cross the blood-brain barrier, as evidenced by the disruption of blood brain barrier integrity in the preclinical multiple sclerosis model, a model that requires the induction of a brain lesion in order to see any disruption to the blood brain barrier integrity.
